In Vitro Properties of Photochemically Pathogen-Reduced Treated Platelet Concentrates Suspended in SSP plus EVER for a 9-Day Storage Period

Background/Case Studies: New platelet additive solutions (PAS) containcompounds that might improve the storage conditions for platelets. Thisstudy assessed in vitro function of platelet concentrate from buffy coat(BC-PC), treated with pathogen reduction technology (PRT) and stored inAS, PAS III M, or SSP+(MacoPharma), that incorporates potassium andmagnesium in relation to PAS III (InterSol). Study Design/Methods: Sev-enteen BC- PC were prepared using five standard BC products pooledtogether with SSP+, in the OrbiSac system (Terumo BCT). The ratio AS: plasma was 65:35 approximately. The units were treated with PRT (Intercept,Cerus Corp.). All products were stored for 9 days under standard bloodbanking conditions. BC-PCs were tested for platelet count, MPV (meanplatelet volume), glucose, lactate, pH (at 22 C), CD62p expression, AnnexinV binding and some biological response modifiers like sCD40L and sCD62p. The assessment of swirling was scored as 1, 2, and 3 (1=no swirling; 2=intermediate; 3=good). Results/Findings: The volume and platelet per unitwere 345±7.8 mL and 3.5±0.2×1011 respectively. pH was maintainedmore than 6.8 at day 9 and swirling remained at good level (score 3) for allunits throughout storage. The residual leucocyte was determined on day 2by flow cytometry (FC),and all BC-PCs had less than 1×106 WBC/unit. The results are shown on the included table. Conclusion: The BC-PCs treatedwith PRT and suspended in SSP+can be stored for 9 days with acceptablecell quality like low glucose metabolism, low levels of spontaneous plateletactivation and others metabolic and biological parameters are adequate.
